Product Intro:
Ibrushil 140mg is a Bruton’s tyrosine kinase (BTK) inhibitor that interrupts malignant B-cell proliferation. This capsule formulation offers a convenient oral treatment option for several hematological malignancies and provides improved progression-free survival rates in patients.

Uses:
Ibrushil is used to treat the following conditions:
  1. Chronic Lymphocytic Leukemia (CLL)
  2. Small Lymphocytic Lymphoma (SLL)
  3. Mantle Cell Lymphoma (MCL)
  4. Waldenström's Macroglobulinemia (WM)
  5. Marginal Zone Lymphoma (MZL)
  6. Chronic Graft-Versus-Host Disease (cGVHD)

Storage Instructions:
  1. Store at room temperature between 20°C to 25°C
  2. Keep away from moisture and direct sunlight
  3. Keep out of reach of children

How it Works (Mechanism of Action):
Ibrutinib is an irreversible inhibitor of Bruton’s tyrosine kinase (BTK), an enzyme crucial for B-cell receptor signaling. By blocking BTK, it inhibits malignant B-cell proliferation and survival, thereby slowing or stopping the growth of cancerous cells in blood and lymphoid tissues.

Side Effects:
Common Side Effects:
  1. Diarrhea
  2. Fatigue
  3. Nausea
  4. Bruising
  5. Muscle or bone pain
  6. Cough
Severe Side Effects:
  1. Bleeding complications
  2. Atrial fibrillation
  3. Hypertension
  4. Cytopenias (low blood counts)
  5. Infections
  6. Second primary malignancies

Dosage (Typical Recommended Dose):
  1. MCL: 560 mg once daily (4 x 140mg capsules)
  2. CLL/SLL/WM/cGVHD: 420 mg once daily (3 x 140mg capsules)
  3. Always follow dosage as prescribed by a healthcare provider

Method of Administration:
  1. Take orally with a glass of water
  2. Swallow capsules whole; do not open, crush, or chew
  3. Can be taken with or without food
  4. Take at the same time each day for best results

Precautions:
  1. Use with caution in patients with bleeding disorders or on anticoagulants
  2. Monitor for infections, especially in immunocompromised patients
  3. Regular ECG monitoring may be required due to risk of atrial fibrillation
  4. Liver and kidney function should be assessed regularly
  5. Avoid in pregnancy and lactation unless clearly necessary

Drug Interactions:
  1. Strong CYP3A4 inhibitors (e.g., ketoconazole, clarithromycin) – may increase Ibrutinib levels
  2. CYP3A4 inducers (e.g., rifampin, phenytoin) – may reduce effectiveness
  3. Anticoagulants and antiplatelet agents – increased risk of bleeding
  4. Avoid grapefruit juice and Seville oranges

Allergies:
  1. Contraindicated in patients with known hypersensitivity to Ibrutinib or any excipients
  2. Monitor for signs of rash, itching, or anaphylactic reaction

Overdose Information:
  1. In case of overdose, monitor closely for signs of toxicity such as severe bleeding or arrhythmia
  2. Provide supportive treatment
  3. No specific antidote exists for Ibrutinib overdose

Missed Dose Instructions:
  1. If a dose is missed, take it as soon as remembered on the same day
  2. Do not take extra doses to make up for a missed dose
  3. Skip the missed dose if it’s almost time for the next scheduled dose

Additional Notes:
  1. Regular blood tests may be needed to monitor blood counts and organ function
  2. Patients should be educated about bleeding risks and infection symptoms
  3. Do not stop taking the medicine without consulting a healthcare provider
