How it Works (Mechanism of Action):
Ibrutinib is a Bruton's Tyrosine Kinase (BTK) inhibitor.
  1. BTK is essential in B-cell receptor signaling that helps malignant B-cells survive and multiply
  2. Ibrutinib forms an irreversible bond with BTK, blocking this signaling
  3. This leads to reduced proliferation and induces cell death in cancerous B-cells

Method of Administration:
  1. Take orally once daily with a full glass of water
  2. Can be taken with or without food
  3. Swallow whole – do not open, crush, or chew the capsule
  4. Take at the same time each day

Precautions:
  1. Monitor for cardiac issues (especially atrial fibrillation)
  2. Use caution in patients with bleeding disorders or on anticoagulants
  3. Avoid use during pregnancy and breastfeeding
  4. Use effective contraception during and for at least 1 month after stopping
  5. Periodic blood pressure checks, ECG, and blood tests are recommended

Drug Interactions:
  1. Avoid strong CYP3A4 inhibitors (e.g., ketoconazole, clarithromycin)
  2. Avoid strong CYP3A4 inducers (e.g., rifampin, carbamazepine)
  3. Interacts with antiplatelets and anticoagulants – increased bleeding risk
  4. Avoid grapefruit juice and Seville oranges

Allergies:
  1. Do not use in patients with known hypersensitivity to Ibrutinib

Overdose Information:

Missed Dose Instructions:
  1. If missed by less than 12 hours, take it as soon as remembered
  2. If more than 12 hours have passed, skip the missed dose
